Biochemical variation during direct organogenesis , regeneration potential viability rate of nodal segments during invitro and TLC screening methods for antioxidant activity of Heena leaves followed by solid phase extracts using F254 plates for scavenging activity

Abstract

 Lawsonia inermis commonly known as Mehandi or Henna related  to family Lythraceae. Lawsonia inermis have antioxidants potential  important role to health and wealth , for example is the implications of the oxidation reaction of reactive oxygen species  in the body that can lead to cardiovascular ailment , anticancer ,  antiaging. Natural antioxidants found  in plants and flavanonols  can come from one or more components such as in Henna leaf, its compound an inhibit the oxidation reaction.The present work was carried out with the objectives to devise suitable media for culture initiation, direct organogenesis  and biochemical estimations of  during differentiation and tlc secreeing method by using F254plates . Murashige and Skoog (1962) medium was used for the present investigation .This medium was fortified  with various amount  of  auxins like Indole acetic acid , Napthalene acetic acid, 2,4-Dichlorophenoxy acetic acid  (0.5,1.0,1.5,2.0  and 3.0mg/l) and cytokinins like Kinetin and Benzylamino purine (0.5,1.0,1.5,2.0 and 3.0mg/l)  individually and in different combinations .The medium containing Indole acetic acid resulted in highest percent induction of shoot induction.. Maximum percent shoot  induction upon cytokinin supplemented media was on BAP (3.0). The higher growth value callus developed from internodal segments was used for biochemical studies. There was decline in the contents of starch whereas the total soluble sugars , reducing sugars and total soluble proteins got increased .The activity of the enzyme alpha  amylase of callus got increased and the activity of other enzymes i. e.  acid invertase and acid peroxidase got decreased with the passage of time. TLC screening  plant extracts revealed that lawsone (naphthoquinone) was found in mother plant , regenerated plants and calli derived  ,direct  oraganogenesis  nodal   segments of Lawsonia inermis. Highest  florescence  and  lawsone content yield (0.154 µg /mg ) was observed in leaves of tissue culture raised plantlets with as compared to mother plant and the nodal explants  and also higher in shoot  develop from nodal segments in direct organogenesis.
